چکیده

Removal of base Y adjacent to the anticodon of yeast phenylalanine tRNA changes the coding properties of tRNAPhe. Phenylalanine tRNA from which base Y has been removed (tRNA:b,“) recognizes UUC better than WU. Codon UUU is read efficiently at higher Mg++ concentration or in the presence of streptomycin. A dipeptide cannot be synthesized with N-acetyl-Phe-tRNAPh” and Phe-tRNA% unless the base Y is also removed from the acetylated PhetRNA. Removal of base Y from the anticodon loop does not affect the ability of phenylalanyl-tRNA?h to form the ternary complex with GTP and the bacterial elongation factor T.

SURVEY AND SUMMARY Decoding the genome: a modi®ed view

Transfer RNA's role in decoding the genome is critical to the accuracy and ef®ciency of protein synthesis. Though modi®ed nucleosides were identi®ed in RNA 50 years ago, only recently has their importance to tRNA's ability to decode cognate and wobble codons become apparent. RNA modi®cations are ubiquitous. To date, some 100 different posttranslational modi®cations have been identi®ed. Modi®cat...

The structure of the human tRNALys3 anticodon bound to the HIV genome is stabilized by modified nucleosides and adjacent mismatch base pairs

Replication of human immunodeficiency virus (HIV) requires base pairing of the reverse transcriptase primer, human tRNA(Lys3), to the viral RNA. Although the major complementary base pairing occurs between the HIV primer binding sequence (PBS) and the tRNA's 3'-terminus, an important discriminatory, secondary contact occurs between the viral A-rich Loop I, 5'-adjacent to the PBS, and the modifi...

Accurate translation of the genetic code depends on tRNA modified nucleosides.

Transfer RNA molecules translate the genetic code by recognizing cognate mRNA codons during protein synthesis. The anticodon wobble at position 34 and the nucleotide immediately 3' to the anticodon triplet at position 37 display a large diversity of modified nucleosides in the tRNAs of all organisms. We show that tRNA species translating 2-fold degenerate codons require a modified U(34) to enab...

Bringing order to translation: the contributions of transfer RNA anticodon-domain modifications.

The biosynthesis of RNA includes its post-transcriptional modifications, and the crucial functions of these modifications have supported their conservation within all three kingdoms. For example, the modifications located within or adjacent to the anticodon of the transfer RNA (tRNA) enhance the accuracy of codon binding, maintain the translational reading frame and enable translocation of the ...

mutant haloarchaeal isoleucine tRNAs a new modified nucleoside found in the anticodon wobble position of Identification and codon reading properties of 5-cyanomethyl uridine,

Most archaea and bacteria use a modified C in the anticodon wobble position of isoleucine tRNA to base pair with A but not with G of the mRNA. This allows the tRNA to read the isoleucine codon AUA without also reading the methionine codon AUG. To understand why a modified C, and not U or modified U, is used to base pair with A, we mutated the C34 in the anticodon of Haloarcula marismortui isole...
